培养pH值对2种饵料硅藻光合生理及脂肪酸组分的效应研究

    Effects of Culturing pH on Photosynthetic Physiology and Fatty Acid Composition of Two Dietary Diatoms

    • 摘要: 为评估培养pH值对饵料硅藻营养价值的影响, 探究引起饵料营养价值变化的pH阈值, 基于培养体系酸碱度调控手段, 以中肋骨条藻(Skeletonema costatum)和威氏海链藻(Thalassiosira weissflogii)为研究对象, 探究不同pH值水平对2种硅藻光合生理和脂肪酸组分的效应。结果表明, 培养pH值在7.0~8.0范围时, 对2种硅藻的生长、相对电子传递速率及脂肪酸占比无显著影响, 而当培养pH值升高至9.0时, 2种硅藻比生长速率及光系统Ⅱ有效光化学效率受到一定程度的抑制, 威氏海链藻可溶性蛋白和可溶性碳水化合物含量增加。中肋骨条藻的饱和脂肪酸占比在不同处理组均超过90%, 在pH值为9.0时二十二碳六烯酸(DHA) 含量最高。当培养pH值在7.0~8.0范围时威氏海链藻不饱和脂肪酸合成受到促进, 在pH值为7.0时其DHA和二十碳五烯酸含量最高。研究结果对水产饵料微藻培养及其营养价值提升具有重要参考价值。

       

      Abstract: To evaluate the impact of pH on the nutritional quality of dietary diatoms and determine the pH threshold at which changes in nutritional value occur, the responses of Skeletonema costatum and Thalassiosira weissflogii to pH variations were studied. The photosynthetic physiology and fatty acid composition of both the species were investigated by manipulating pH in the culturing system. Results show that, within the pH range of 7.0 to 8.0, pH had no significant effects on the growth, relative electron transfer rate, and fatty acid composition of both diatoms. However, when culturing pH increased to 9.0, the growth rate and effective quantum yield of photosystem II in both diatoms were moderately inhibited, with an increase in soluble protein and soluble carbohydrate in T. weissflogii. The saturated fatty acid content in S. costatum exceeded 90% of the total content of fatty acid content in all treatment groups, with the highest content of docosahexaenooic acid (DHA) at pH 9.0. The synthesis of unsaturated fatty acids in T. weissflogii was promoted in the pH range of 7.0 to 8.0, with the highest DHA and eicosapentaenoic acid content observed at pH 7.0. These findings offer important insights for cultivation guidance and improving the nutritional quality of dietary microalgae.
